The procedure described here can be adapted to the enrichment of other repetitive elements, based on the use of restriction enzymes that do not cut into the repeat of interest. The latter can be combined with specific labeling for single-molecule analysis of replicating DNA or for long-read sequencing analysis of telomeric repeats. A smaller-scale version of the protocol that involves one round of digestion and purification requires 200 µg of DNA and enriches telomeres ~50-fold in 4 d is also provided. No special skills are required for the enrichment procedure, while some assistance is needed in harvesting a large number of plates in a timely fashion at the beginning of the procedure. The purified material is suitable for single-molecule analysis of telomere structure, visualizing telomere replication and recombination intermediates by electron microscopy or performing molecular combing at telomeric repeats. Around 2 mg of genomic DNA is required, and the procedure lasts 5–6 d and yields preparations enriched >800-fold in telomeres. ![]() The procedure consists of two successive rounds of digestion with frequently cutting restriction enzymes followed by size fractionation. Here we provide a detailed protocol for the enrichment of telomeric repeats from mouse and human cells.

If you don’t mind me prattling on further - I’m having a moment today! - I feel staunchly that one way we can make cooking feel like more than just a list of endless tasks to complete, even though it’s definitely often that, and more than about pleasing the people who it feeds, elusive as that can be with kids or really any time validation comes from the outside, is to focus on how it feels to make something that tastes, smells, and feels phenomenal and this, for me, is very much that. No seriously: It’s like a thick comforter on a shivery rainy day, it tastes the way the first hot cup of coffee of the season feels in your hands, and it smells the way you dream your home will when you open the door after an exhausting day, or so seems to be the reaction from my two most opinionated dinner guests. Just imagine how much salivating will go on when you fire up a five-burner grill and load it up with brisket, buttered vegetables, and other delectables. Sure, particular food just naturally tastes better when prepared on an outdoor grill, but the smells that grills put out get your guests salivating before they put that first bite into their mouths. Marinades, sauces, butter, and so on float through the air, making everyone extra hungry. What are the Advantages/Benefits of 5 Burner Gas Grills?Īccording to Napoleon much of the reason barbecued food tastes better to us than food prepared indoors is the smells associated with smoking and cooking the food. And there are also many features to consider. Your budget may dictate that you purchase a free-standing grill. Outdoor kitchens and the grill heads they require would separate you from quite a bit of cash. Get the appropriate grill instead of adapting an ill-suited grill. But do you have an outdoor kitchen? Do you plan to build one? A five-burner grill head would be what gets dropped into your outdoor space. At least you intend to cook a lot of food at once. Since you are in the market for a five-burner grill, you obviously intend to cook for a lot of people. 6 Best 5 Burner Gas Grill: The Verdict How to Choose the Best 5 Burner Gas Grill 12/9/2023 0 Comments Artful ly![]() Right now, some of the most popular feature requests include selling memberships and subscriptions, patron house holding, and print at home tickets. That idea got a ton of votes in the forum so we found the money and the time to make it happen. For example, an integration with MailChimp was a suggestion we heard at the beginning of our private beta period. We take those requests really seriously and the features with the most votes in the Feature Request Forum are the things we prioritize building. “Community Driven Design” is as core principle for us–we’ve hosted focus groups all over the US and have hundreds of participants our public Feature Request Forum. That’s what we started from, but we’ve also been actively listening to artists all along. They were tired of vendor lock-ins that hijacked their data, outrageous licensing fees (some systems costs tens of thousands of dollars a year), and wacky protocols for entering the same data in four or five different places. They wanted a patron management system that was smart about social media, had robust searching and segmenting options, and gave the little organizations the power to do targeted marketing and donor cultivation just like the big guys. Arts groups wanted something built for the arts, no re-purposed from some other industry. We heard a need for a platform that puts tickets, donations, marketing, and contact management all in one place and is affordable, intuitive, flexible. ![]() Juneau-Vogel: Five years ago, this project was inspired by some pain points our members and other artists were voicing. Both financially speaking and in terms of optimizing audience outreach. If you also work as a teaching artist, you can use Artful.ly to register participants in any workshops you may be running. If you run an IndieGoGo or RocketHub crowdfunding campaign, all the donor data from the campaign will get imported into the Artful.ly account. If he or she is tracking some of this data already in a spreadsheet, it’s easy to import into Artful.ly, and you can sync MailChimp email newsletter subscriber list to Artful.ly too. If the filmmaker has a nonprofit or is fiscally sponsored, you can use Artful.ly to take donations and keep track of donors. He or she can log volunteer hours, tag individuals for quick searches, and export contact lists for mailings. We’ve also seen individual artists use Artful.ly to take RSVPs and sell tickets for gallery shows, workshops, and meetings.Īn individual filmmaker could use Artful.ly’s CRM tools to keep track of contact info for production staff, actors, and volunteers. Individual artists especially like MailChimp integration which puts newsletter subscription info alongside donor info. Generally, individual artists use Artful.ly for the CRM features - managing contact info, tracking interactions with fans, donor cultivation notes, logging donations, etc. But yes, there are several individual artists using Artful.ly too. We have a few festivals and presenting venues. Juneau-Vogel: The majority of current Artful.ly users are small theaters, dance groups, music ensembles, and other performing arts groups. ![]() Filmmaker spoke with Selena Juneau-Vogel, program director for Artful.ly, about how filmmakers can make the most of the new software.įilmmaker: I noticed that many of Artful.ly’s beta testers were theater companies and festivals, but do you feel it’s applicable to individuals? How could a filmmaker, for instance, make the most out of its services? There are no sign-up fees or contracts, however, in the interest of an event, ticket buyers will be charged $2 per purchase in addition to credit card processing. Among the service’s offerings is a MailChimp integration that directly manages and catalogues email outreach, and a ticketing and fundraising apparatus that can be embedded onto websites. Aiming to give independent artists and arts organizations the same marketing power as their bigwig counterparts, the non-profit Fractured Atlas recently unveiled Artful.ly, a web-based software system that streamlines audience interactions. 12/9/2023 0 Comments Artrage gallery syracuseThis work has been much discussed over the decades. At the SU Art Galleries in the Shaffer Art gallery you can see several of Fraser's works included a bronze model of his famous End of the Trail, sculpted for the 1915 Panama-Pacific Exhibition. Her representation of the Onondaga goes beyond the (then) popular notion of the 'noble savage," to include them as full community partners - a partnership then denied to both Indians and all American women.Ĭorbett's contemporary and fellow Augustus Saint-Gaudens and Art Students League student James Earle Fraser (1876-1953) - who created some of the most lasting images of the Western Indian - is also well represented in Syracuse. I've already written about her magnificent Kirkpatrick Monument recently restored in Washington Park. She studied sculpture with Augustus Saint-Gaudens at the Art Students League in New York later studied at the Ecole des Beaux-Arts in Paris (1898-99), before creating several impressive bronze monuments in her hometown, and then establishing herself in New York. I think it significant that the two greater works of art, that are also the most heroic representation of Indians, are by two notable women sculptors with ties to Syracuse - Gail Sherman Corbett (1872-1951) and Luise Kaish (b. Microsoft initially aimed to have Windows 10 installed on over one billion devices within three years of its release that goal was ultimately reached almost five years after release on March 16, 2020, and Windows 10 is now the most used version in virtually all countries. However, media outlets have been critical of the changes to operating system behaviors, including mandatory update installation, privacy concerns over data collection performed by the OS for Microsoft and its partners, and adware-like tactics used to promote the operating system on its release. So how can we tell if a chemical change has occurred? 1. Indications of a Chemical Reactionīased on its definition, a chemical reaction occurs when a substance undergoes a chemical change. While some physical changes are easily reversible, such as re-freezing melted ice, reversing a chemical change requires another chemical reaction. This process changes the chemical composition of the original substance. When an iron nail comes into contact with water and is then exposed to the air, it rusts, forming a brown-red substance. Once melted or boiled, the water may be in a different form (solid ice or gaseous water vapor), but it is still water, H 2O, and it still has the same chemical composition.Ī chemical change happens when a substance’s chemical identity changes. ![]() ![]() For example, freezing or boiling water is a physical change. In a physical change, a compound’s shape may change, but its chemical identity will not. Let’s looks at some chemical change examples, and chemical reaction examples. Chemical equations express chemical reactions in terms of symbols and chemical formulas of the compounds involved in reactions. So what is a chemical reaction? What is a chemical change, and what is a physical change? A chemical reaction occurs when compounds or substances undergo a chemical change to form different compounds or substances. 12/9/2023 0 Comments Snark tuner for banjoyou might like it and its very conveniently mounted, with that adaptor, right on the banjo hooks, for easy reach. No offense is meant and I dont really change my posting style to suit what folks 'like'. Im sorry you dont like my posts but to be honest Im just being me and responding to what i read. Why is it that disagreeing with anyone on a forum is now being 'snarky' ? I just disagreed and said i wouldn't think it good advice to tell folks to throw away perfectly good tuners simply because you prefer a 'Korg'. ![]() You also said 'if anyone wants to use that tuner."more power to them', which is an oft used euphemism which implies 'snarky'. I didnt need to leave out anything as your reply was right there to read. You then said you prefer to use your ears, then said you used an electronic tuner yourself, which completely contradicted the earlier remark. I thought that was bad advice and completely subjective. You stated that its a 'good idea to discard it (electronic tuner) altogether", which was a statement aimed at those who do use them. I wasn't intending to be snarky, but to point out that an electronic tuner is a tool, just like a capo and picks. I used to really like your comments, but lately you are becoming a little "snark-y." (see, anyone can use an emoto-con) Why do you want to distort what a fellow poster says and add things that he did not say and twist around things that he did say just to make yourself look like some great authority? You also left out my last two sentences. That's funny, but I didn't say anything about straps or capos or any other devices. The only difference between a Snark and your devices is the Snark mounts conveniently on your banjo pot assembly and your two wont. ![]() You said you used a tuner and a tuning fork, so you are not just 'trusting you ears', as you stated. I think telling folks to discard these tuners is excessive and bad advice, they should also toss away straps and sit down to play and throw away a capo too. In certain situations where ambient noise is a real issue, tuners are invaluable for tuning, when everyone can't hear and know if they are in tune or not, tuning to each other is blind leading the blind. There are situations when tuning to other instruments is a recipe for disaster. I have an excellent ear, but not perfect pitch. If one likes and uses the electronic tuners, then more power to them. I had rather use my ear than depend on what some electronic device is telling me. I prefer to tune to another instrument or even a tuning fork. I do occasionally use a Korg hand held tuner, especially when changing strings. I don't like electronic tuners, especially the kind that clips on the peg head. A good reason to just discard it altogether. 12/8/2023 0 Comments Turn off instagram login activity![]() Whether you log in using a dedicated Instagram account, or by using Facebook, your Instagram profile is already secured with a password. But it does share your data with third parties - including advertisers, researchers, and law enforcement agencies - when it is deemed necessary. It also says that Instagram imposes "strict restrictions" on how its partners can use and disclose the data it provides. One thing to note about Instagram's official privacy policy is that it strictly states it does not sell - and never will - user data. The vast majority of that is then used to serve you personalized or "targeted" ads based on what Instagram thinks you like. This not only includes the information you submit when joining Instagram, but also your interests and browsing history. Our report found that Instagram shares up to 79% of the personal user data that it collects. One study, conducted earlier this year, labeled Instagram one of the most "invasive" apps for data sharing. In other words, it is not exactly the most conservative when it comes to looking after your data. When it comes to data collection, Instagram's privacy policies are similar to those of Facebook, since both platforms are owned by the same company - now called Meta. Beyond its default settings, Instagram offers a number of features - including two-factor authentication - that are designed to bolster account security. However, the security of your account mostly depends on you - how well you protect your account and keep your login details safe. Instagram itself is a secure platform, so the chances of it being hacked are slim. ![]() In this guide, we'll cover all the things you can do to make your Instagram account more secure and step up your privacy.
